01Do I need to have done Masterclass 1 first?

You don't need to have attended it, but you do need the essentials: the evolution of money, centralised vs decentralised systems, tokenisation, and how DeFi and self-custody work. You don't need to be an expert. If those topics are new to you, start at defimasterclass.app first, because Masterclass 2 will feel overwhelming without them.
